A culture is a group of microorganisms that has been cultivated in a lab. A mixed culture comprises two or more different types of bacteria while a pure culture only contains one type. While a mixed culture comprises a variety of microorganisms, a pure culture only contains one organism.
In order to adequately streak a plate for isolation, the organism(s) must be dispersed via the inoculating loop until single colonies appear. Each colony is made up of a collection of cells that develop from a single bacterial cell through cell division. Each mixed separated colony thus represents a pure bacterial culture.
