Beadle and Tatum experimentation on Neurospora crassa (A red bread mold microorganism) with haploid genes and its prototrophic meaning that it could grow on a minimal medium and they have ability to synthesize the amino acids and vitamins necessary for growth

They first placed the Ascospores in the complete media and knowingly exposed the spores to X-Rays which lead to the mutations in the DNA. After the exposure to the x-rays some cells still had the ability to grow but some cells became auxotrophic meaning they no longer grow in the minimal medium but they had the ability to grow in the complete medium

They then placed the spores in different vials containing minimal medium to see the ability of the  spores to grow after experimenting on 2000 samples to identify 3 mutations involved in the synthesis of pyridoxine (B6), Thiamine (B1), Para-aminobenzoic acid

They concluded that the inability of a mutant to produce to produce vitamin B6 such mutant called pyridoxine mutant differentiated by a single gene which they had mutant in spores via X-Rays.
