What is DNA MTase activity and PEC assay?

  • Based on the exciton energy transfer (EET) effect and site-specific cleavage of restriction endonuclease, a highly sensitive photoelectrochemical (PEC) test for MTase activity and inhibitor screening was created.
  • The test was created by fusing CdSe quantum dots (QD) with probe DNA that was tagged with Au nanoparticles (NPs).
  • The photocurrent signal was drastically reduced as a result of the potent EET effect between Au NPs and CdSe QDs.
  • A sensing region on the pDNA allowed it to recognise the target DNA (tDNA) and hybridise with it to create a DNA duplex.
  • The photocurrent signal was restored once the EET was broken by the site-specific cleavage of HpaII, which allowed the DNA duplex to be broken and Au NPs to be released.
